SUBJECT

: Project MKULTRA, Subproject No. 135, :

Change of Facility in which Work is Performed.

1. The ori ginal purpose of this subproject was to evaluated

a number of candidate chemical a gent s in facilities and under the
direction of individuals who, if later developed, became unavailable
for this purpose d\t a to unforesee n circumstances.

2. In the meantime, the funds for this subproject were

transferred to the cutou t in antici pation of the commencement of
the work,

3. In view of the security difficulties involved in the actual
return of funds from the cu tout and also since another s ubpro ject
(#91) was being served by the same cutout, and furthermore, since

subproject #91 was ca pable of providing its services on these

chemical agents of interest an d durin g th e s ame fiscal year, it has
been decided to divert the wor k i n tended for sub project #135 to the
facilities of subproject #91. No significant change in funding or
variation in fiscal year is concerned in this matter.

1. The pur jposezqOE^^ybprPj eet is to provide fu^ds to carry out
a research program involvin g utilizat ion of human vol unteer subjects.

2 . This prog ra m is designed to ex ploit clinical and laboratory Z .Z1_„..' Z.;Z^ZJ_„Z
facilities and professional knowledge in providing answers to specific

questions of direct interest to the Agency. The nature of the investi-
gations to be carried out encompasses pharmacological and clinical

testing in human volunteers of chemicals, biochemica ls, and biologically
active materials und er c ontrolled conditions. For exa mple, human
patch testing or human blood group determinations will be mounted upon

setting up a suitably designed experiment,

This individual will serve as project

leader and coordinate all ph ases of th e r ese ar ch p rogram, select and

provide clinicians to conduct the exp er iments, assist in planning the

experimental design and protocol, and supervise the preparation of data

some of the investigations that will be carried out.

PROTOCOL

OBJECTIVE :

To determine whether an unknown compound, either chemical
or biological, is capable of eliciting a reaction when applied to picall y^
to the skin, either locally or systemically, or both.

BACKGROUND :

r If the skin is able to deal with the irritanLcompound, die reaction
is physiological or normal. However, if the action of the irritant is ex-
cessive , the reaction is patholo gical or abnormal and irritation results .
Irritation is an extreme reaction of tissues to an insult or injury and is
best characterized as incipient inflammation.' The inflammatory process
may vary from a b arely p erceptible h y peremi a, to edema formation and
vesieulation and finally to a suppurative process . - - - - , - :

A primary irritant may be defined as a substance p roducin g an
injur y on fi rst c ontact . T he resultant injury will depend on the nature
of the Irritant, concentration of the irritant, and the total elapsed time
of exposure. Primary irritation of the skin is' usually measured Exper-
imentally b ythe patch test technique on abrad ed or intact skin of the ex-
perimental animal, for example, rabbits, guinea pigs or man.

screen all compounds to be tested to obviate exposing man to severely
sens itiz in g substances . G r anted, in t h e c ase of compounds which may
be only mildly sensitizing or in which small animal studies are unable

Animal experimentation data should include acute , subacute and

chronic dermal toxicity experimentation that will allow some logical
.. assessment of the local and systemic toxicity of substances to be applied

topically to man in the pro per amounts, conce ntration and frequency of
applic ation that may be tolerated. ;

PROCEDURE :

For pilo t s tudies, human testing should employ 20 test subjects
per compound tested. Prior ^ experimental testing, maximum efforts
must he made to minimize the r isk o f incurring unf o rs e eahle sens i tivi-
ties and untoward res ponses" by establishing a base line profile that will
encompass the foUpwing:

1 . Complete medical histories with emphasis on known sensi-
tivities and^dfu g idiosyncraciesTTl

2 . Preliminary physicial examination -

3. Hematological evaluation ( complete blood count )

4. Urinalysis ( microscopic )

5. Radiolog ical screening ( chest and long bone )

Acute dermal toxicity. ( Single exposure or primary irritation )
This will utilize the patch test techniq ue on intact skin on a min-
imum of five subjects per test compound . ,

Methodolog y will c onsist of introducing under a one in ch patch

0 . 5 ml (liquid) or 0.5 g m (solid or semi- soli d) of test substance and
securing the pitch in place with non- sensitizing adhesive.

After 24 hours, the patch is remoyed and readings made on the
resultant reactions evaluated on the basis of the following scores :

final score represents the average of 24-72 hour readings.
